Ethyl cyanohydroxyiminoacetate (oxyma) is the oxime of ethyl cyanoacetate and finds use as an additive for carbodiimides, such as dicyclohexylcarbodiimide (DCC) in peptide synthesis. It acts as a neutralizing reagent for the basicity or nucleophilicity of the DCC due to its pronounced acidity (pKa 4.60) and suppresses base catalyzed side reactions, in particular racemization. OxymaPure®, an ideal additive used with carbodiimide coupling methodology, for amide bond formation. The 2-cyano-2-(hydroxyimino)acetate displays a remarkable capacity to suppress racemization and an impressive coupling efficiency in both automated and manual synthesis. OxymaPure® derived products are extensively adopted in pharmaceutical platform for the synthesis of various scaffolds such as Weinreb Amides; Oligobenzamides; ester bond formation and more. Oxyma Pure is a non-explosive alternative to HOBt. When used in place of HOBt in carbodiimide-mediated coupling reactions it provides products in higher yields with less racemization. OxymaPure® used with DIC produces peptides with increased yield and decreased epimerization when used as an alternative to HOBt.1 This safe, non-explosive auxiliary nucleophile works with carbodiimide coupling strategies to provide the best results for a peptide synthesis. Additionally, the use of DIC/Oxyma avoids side reactions associated with high levels of base (≥ 1 equiv. DIEA) using onium salt methods such as HBTU/DIEA.
