HCTU is a novel aminium-based coupling reagent and analogue to HBTU and HATU. In comparative studies, HCTU was found to give superior results to other coupling reagents in the synthesis of difficult and long peptides. HCTU is also reported to be less allergenic than other coupling reagents.
Advantages of HCTU:
Fast: The coupling step could be complete in as short as 1 minute. For synthesis of a 10-mer peptide with double coupling and double deprotection, the cycle time could be about 14 minutes and the total synthesis could be finished in around 2 and half hours.
Low Cost: Comparing to HATU, the similar effecient coupling reagent, the cost for HCTU is about one quarter.
High Stability: The solution of HCTU in DMF could last for a few weeks without significant degradation, enable it to be in the pre-prepared solution for use on synthesizers.
Good Solubility: HCTU displays a better solubility than HATU and HBTU in DMF, which allows the coupling reaction to be performed at higher concentrations and thus improves the total efficiency of the peptide synthesis.
High Yield and Purity of Product: For synthesis of difficult, highly hindered, cyclic and long peptides, HCTU showed high efficiency to deliver higher yield and purity of the product than most of other coupling reagents.
